Results: Hypoxic stress suppressed a higher MW form of CTCF by de-SUMOylation associated with lysine 74 and lysine 689 residues, resulting in significantly inhibited Pax6 expression. Conclusion: Hypoxic stress induces de-SUMOylation of CTCF to functionally regulate CTCF activity. Significance: CTCF plays important roles in growth factor/stress-regulated cell fates through post-translational modulation to control gene expression.

Activation of Oxidative Stress-Regulated Bcl-3 Suppresses CTCF in Corneal Epithelial Cells

Epigenetic factor CTCF (CCCTC binding factor) plays important roles in genetic controls of the cell fate. Previous studies found in corneal epithelial cells that CTCF is regulated by epidermal growth factor (EGF) through activation of NF-κB p65/p50. It also found that CTCF is suppressed in ultraviolet (UV) stress-induced corneal epithelial cells. However, it is still unknown how UV stress down-...

CTCF-Mediated and Pax6-Associated Gene Expression in Corneal Epithelial Cell-Specific Differentiation

BACKGROUND The purpose of the study is to elicit the epigenetic mechanism involving CCCTC binding factor (CTCF)-mediated chromatin remodeling that regulates PAX6 gene interaction with differentiation-associated genes to control corneal epithelial differentiation. METHODS Cell cycle progression and specific keratin expressions were measured to monitor changes of differentiation-induced primary...

Effect of CTCF-binding motif on regulation of PAX6 transcription.

PURPOSE Previous studies indicate that the CCCTC binding transcription factor (CTCF) regulates homeobox PAX6 gene transcription in corneal epithelial cells. In the present study, the effect was investigated of CTCF activity on PAX6 transcription through interaction with five essential motifs located in an 80-bp region upstream from the PAX6 P0 promoter. METHODS An electrophoretic mobility shi...

EGF-induced Corneal Epithelial Wound Healing Though NF-κB Subtype- Regulated CTCF Activation

Capsule Background: To study NF-κB-activated CTCF in EGF-induced wound healing. Results: Corneal epithelial wound healing was significantly impaired because of lacking CTCF activity in NFκB-p50 mice. EGF-induced NFκB activation regulated CTCF by interacting with CTCF-promoter to increases motility, migration and wound healing. Conclusion: CTCF is a NFκB-p50-interactive target in EGF-induced cor...
